The fluorescence peaks of the two IR dyes used with the Global IR
System are separated by over 110 nm. Two independent
excitation/detection channels are used to eliminate errors due to
fluorescence overlap. Each channel is optically tuned to measure
fluorescence from only one of the IR dyes.
One laser diode emits radiation at 680 nm, and the other at 780 nm.
Peak power rating is 30 mW. Each laser is electronically pulsed to
optimize the signal-to-noise ratio and provide temporal discrimination
against any background fluorescence from the gel plates.
Laser diode technology provides high reliability, low power
consumption (<0.2 W), low noise, low cost, and small weight and
size. Because of their small size, the diodes are mounted directly to

The laser radiation is collimated, filtered, and focused to an elliptical
spot size of 30 ?m x 50 ?m. A small spot size allows better resolution
of closely spaced bands as they pass the detection window.
Radiation emitted by the excited fluorophores is collected by one of
two microscope detectors. Radiation is filtered by a bandpass filter
to assure that fluorescence from only one of the IR dyes is measured.
The filter also removes scattered radiation and Raman scattering due
